P. falciparum lacks the de novo pathway for purine nucleotide synthesis and depends on an essential salvage pathway, a key drug target, for survival. Enzymes of this pathway that have been under study are hypoxanthine guanine phosphoribosyltransferase (HGPRT), adenylosuccinate synthetase (ADSS), adenylosuccinate lyase (ASL), GMP synthetase (GMPS), AMP deaminase (AMPD) and IMP-specific nucleotidase I (ISNI). We have also established the presence of an essential, metabolic proof-reading machinery in the parasite.
