Wild-type p53 is regulated by coactivators like PC4 and p300. PC4 strongly interacts with p53 and plays a key role in p53 mediated apoptosis. Both wild type and mutant, is a bona fide inducer of p300 autoacetylation, thereby regulating the epigenetic landscape. Consistent with this, p53 activity is compromised in oral cancer either through mutations or changes in the status of p53 modulators. Missense mutations in TP53 can contribute to tumorigenesis through gain-of-function of biochemical and biological properties to contribute to oral oncogenesis.
